In agarose gels under submerged conditions, is the most efficient way to separate nucleic acids (DNA/RNA) fragments. In this mechanism, the entire gel is submerged below the surface of the buffer. In order to make sample wells in gel, suitable lot  formers (also known as ‘Comb’) is place in the gel platform during gel casting. After the gel is set, the comb is taken and rectangular wells are obtained.
